Effect of 2ccPA on mRNA expression and production of MMP-1, -3, and -13. To measure mRNA expression, synoviocytes (A) and SW1353 cells (B) were cultured with 1 or 10 μM 2ccPA for 24 h in the presence or absence of 10 ng/mL IL-1β. mRNA levels were determined by quantitative real-time PCR. Data represent the mean ± SE of triplicate independent experiments (*P < 0.05 and **P < 0.01 vs. vehicle-treated controls). To measure MMPs in the culture media of synoviocytes (A) and SW1353 cells (B), cells were cultured with 1, 3, or 10 μM 2ccPA in the presence of 10 ng/mL IL-1β. After 24 h incubation, culture media were collected and the concentration of each MMP was measured by ELISA.
