Concentration-dependent inhibitory effects of Ro-256981 in the arthritis pain model. Administration of Ro-256981 into the CeA 5–6 h postinduction of arthritis inhibited the responses of CeA neurons (n = 6–8 neurons for each concentration) to normally innocuous and noxious stimuli but not background activity. Numbers refer to the concentrations in the microdialysis probe (MD). Ro-256981 was administered into the CeA by microdialysis for 15 min. All averaged values are given as the mean ± SEM and are expressed as percent of predrug control values (in ACSF; set to 100%). The following formula was used for non-linear regression analysis to obtain concentration-response curves and calculate IC50 values: y = A+(B-A)/[1+(10C/10X)D], where A is the bottom plateau, B top plateau, C = log(IC50), and D is the slope coefficient (Prism 3.0, GraphPad Software). * P < 0.05, ** P < 0.01, *** P < 0.001, compared to predrug control values (Dunnett's multiple comparison test).
