Schematic of I228M mutation. Sequence alignment of DI/S4 from human sodium channels. The charge-conserved substitution in DI/S4 replaces a highly conserved isoleucine residue at the cytoplasmic end of the S4 helix. I228 is conserved in all human sodium channels except for NaV1.9 which has a conservative substitution, valine, at the corresponding position.
