Schematic representation of the facilitation of KOR/MOR heterodimerization by E2. Biochemical and behavioral experiments suggest that ERs work cooperatively to increase KOR/MOR expression. We postulate that E2 triggers a signaling complex containing one or multiple ERs, which via an unknown mechanism enhances the formation of KOR/MOR heterodimers and thereby creates the sex difference in opioid actions.
